Sulfide metabolites in the diabetes and pancreatic cancer
Dr. Shen began to direct/run analytical Redox Biology Sub-Core in COBRE (PI: Dr. Chris Kevil ) since Feb 2019, providing the services for redox signaling molecules measurements, such as reactive oxygen species, reactive nitrogen species, reactive sulfur species, etc.
Dr. Shen was invited to attend 2019 Proteomics Core Directors and Staff Symposium in Little Rock, Arkansas, February 21-22, 2019.
During postdoctoral training in Dr. Kevil’s lab, Dr. Shen has developed monobromobimane (MBB) method to measure free H2S and sulfide pools (acid-labile sulfide and bound sulfane sulfur) by HPLC and mass spectrometry. This very sensitive, specific, and useful method has advanced the H2S biology field and universally accepted to assess H2S bioavailability.
Dr. Shen’s research is focused on the understanding of sulfide metabolites in the diabetes and pancreatic cancer by using proteomics technologies and redox analytical approaches.
Hydrogen sulfide therapy in diabetes type 1:
Type 1 diabetes (T1D) is a T cell-mediated autoimmune disease in children and young adults, characterized by the destruction of insulin-producing β cells in the pancreatic islets. Different treatments have been applied for diabetes, including decreasing blood glucose and lipid content. However, those also will result in the development of complications, emphasizing the need for novel approaches. Hydrogen sulfide (H2S) is a critical regulator in cellular metabolism, immunological responses, and inflammatory responses. In recent years, accumulating evidence have shown that H2S regulated the function of β cells as well as that H2S possibly regulates injury responses of β cells. Similarly, H2S can also modulate vascular endothelial function. My lab focuses on the understanding of H2S biology in the diabetic development by using NOD mice model and mass spectrometry analysis.
Emerging role of hydrogen sulfide in pancreatic cancer:
Pancreatic cancer is one of the most aggressive human tumors due to its high potential of local invasion and metastasis. Despite the advances in therapeutic strategies including surgical techniques as well as local and systemic adjuvant therapies, the overall survival in patients with pancreatic cancer remains dismal and has not improved substantially over the past 30 years. Median survival from diagnosis is typically around 3 to 6 months, and the 5-year survival rate is less than 5%. Identifying early marker of pancreatic cancer is a priority for oncology researcher as early detection of the disease is critical to improving patient survival rates. The objective of this project is to identify the hydrogen sulfide-related biomarker using FFPE tissue from patients with pancreatic cancer and mass spectrometry analysis.
