Abuse of opioid drugs is a growing public health problem in the United States. In addition to classical opiates such as heroin and morphine, there have been dramatic increases in the nonmedical use of prescription opioids, such as oxycodone and hydrocodone. Recently, the correlation between opioid analgesics and alternations in brain structure, such as leukoencephalopathy, axon demyelination, and white matter lesions, has been demonstrated in patients with a history of long-term use of prescription opioids. Ours and other's studies indicate that the opioids alter gene expression and signaling pathways in different regions of the brain. The research of our group is focused on mechanisms of opioid-induced neuronal degeneration using animal and cell culture models as well as biomedical methods. Data from our studies suggest that aberrant translation mechanism, called integrated stress response, contributes to opioid-induced neuronal degeneration using blood samples. Such method could be used to detect toxic side effects of opioids and other medications and monitor progression of aging and neurodegenerative diseases. In addition, in collaboration with computer scientists from LSU-S, we analyze trends in drug abuse among hospital visitors using large data analysis.
